Section II: WRBU Sand Fly Mounting Protocol
Maintaining the data chain while taking a DNA sample, clearing, mounting, labeling and finally making a determination is extremely important. The process will likely take multiple weeks to complete and often involve hundreds of specimens. In order to avoid confusion and mistakes, it is important to maintain accurate and up to date lab notes.
1. For specimens stored in alcohol, remove specimen from microtube and place in glass crucible; replace any existing ETH with 95% ETH to increase concentration and wash specimen.
2. For other specimens...
3. Record specimen number, sex and condition in lab notebook prior to mounting.
