Taking the DNA Sample
The two mid legs are the preferred choice to be used for DNA barcoding. If missing or damaged, 1st pair of legs, or as last resort, 3rd pair of legs, may be used instead. To avoid sample contamination, wear latex gloves while taking DNA samples.
1. Using a pair of forceps, remove the two midlegs and place in a microtube well filled with ETH. We typically use 96-well microplates (BioQuip).
2. Note the well number containing the sample in the lab notebook.
3. Clean forceps with ETH before pulling the next sample.
